1. What is RBC phenotyping? The phenotype of RBCs (RBC phenotyping) refers to determining the type of antigens present on the RBC. The ABO/Rh type in the ‘type and screen’ is performed on all patients requiring transfusions. However, an extended antigen phenotype may also be performed.
How is Rh typing done?
If you don’t have Rh factor in your blood, you are Rh negative. If you do have Rh factor in your blood, you are Rh positive. Most people are Rh positive. But Rh negative blood types can be passed down from parents to children.
How is Rh serum prepared?
A sample of the mother’s blood is centrifuged to separate her plasma (which contains the antibody) from her red blood cells. The plasma is then serially diluted and combined with reagent Rh-antigen–positive red cells. Agglutination or lack of agglutination is visually assessed.

The Anti-D reagents: Anti-D, Anti-D (PK 1), Anti-D (PK 2), are used to determine the Rh type. They are used to detect the presence of the D (Rh) antigen on the surface of human red blood cells. The Anti-C, Anti-E, Anti-c, Anti-e, and Anti-K are used for Rh-Kell phenotyping of human red blood cells.
